    Abstract

    Keratinocytes and fibroblasts are the major cell types in the epidermis and the dermis, respectively. Since both cells play important roles in wound healing, compounds that regulate their proliferation are expected to be beneficial to epidermal health. We have previously shown that the extract of Emblica officinalis, which is a well-known herb in Ayurvedic traditional medicine, increases the proliferation of keratinocytes. Here we purified a compound that increased cell proliferation from E. officinalis extract. Through liquid chromatography electrospray ionization tandem mass spectrometry and NMR techniques, the compound was determined to be 1,6-digalloyl glucose. Consistent with this, tannic acid, a type of polygalloyl glucose, also increased cell proliferation. We further found that E. officinalis extract increased the proliferation of fibroblasts as well and showed that it increased cell proliferation at least partly through upregulating the expression of periostin. These findings revealed the mechanism by which E. officinalis extract regulates cell proliferation.

    Abstract

    Mammalian cells undergo irreversible proliferation arrest when exposed to stresses, a phenomenon termed cellular senescence. Various types of stress induce cellular senescence; nonetheless, senescent cells show similar phenotypes overall. Thus, cells undergo cellular senescence through the similar mechanisms, regardless of the type of stress encountered. Here we aimed to reveal the mechanisms underlying cellular senescence. We have previously shown that lamin b receptor (LBR), which is a protein that regulates heterochromatin organization, was downregulated in senescent cells, and downregulation of LBR induced cellular senescence. Additionally, we have shown that downregulation of protein synthesis effectively suppressed cellular senescence. Thereby, chromatin organization and protein synthesis are implicated in the regulation of cellular senescence. We examined the roles of them in cellular senescence and found that protein synthesis was upregulated during the induction of cellular senescence, and upregulated protein synthesis caused disturbed proteostasis that led to the decreased function of LBR. Furthermore, we showed that decreased LBR function induced cellular senescence through altered chromatin organization and increased genome instability. Importantly, these findings revealed a link between protein synthesis and chromatin organization and accounted for the phenotypes of senescent cells which show disturbed proteostasis, altered chromatin organization, and increased genome instability. Our findings provided the general model for the mechanisms of cellular senescence.

    Mammalian somatic cells undergo terminal proliferation arrest after a limited number of cell divisions, a phenomenon termed cellular senescence. However, cells acquire the ability to proliferate infinitely (cellular immortalization) through multiple genetic alterations. Inactivation of tumor suppressor genes such as p53, RB and p16 is important for cellular immortalization, although additional molecular alterations are required for cellular immortalization to occur. Here, we aimed to gain insights into these molecular alterations. Given that cellular immortalization is the escape of cells from cellular senescence, genes that regulate cellular senescence are likely to be involved in cellular immortalization. Because senescent cells show altered heterochromatin organization, we investigated the implications of lamin A/C, lamin B1 and lamin B receptor (LBR), which regulate heterochromatin organization, in cellular immortalization. We employed human immortalized cell lines, KMST-6 and SUSM-1, and found that expression of LBR was upregulated upon cellular immortalization and downregulated upon cellular senescence. In addition, knockdown of LBR induced cellular senescence with altered chromatin configuration. Additionally, enforced expression of LBR increased cell proliferation likely through suppression of genome instability in human primary fibroblasts that expressed the simian virus 40 large T antigen (TAg), which inactivates p53 and RB. Furthermore, expression of TAg or knockdown of p53 led to upregulated LBR expression. These observations suggested that expression of LBR might be upregulated to suppress genome instability in TAg-expressing cells, and, consequently, its upregulated expression assisted the proliferation of TAg-expressing cells (i.e. p53/RB-defective cells). Our findings suggest a crucial role for LBR in the process of cellular immortalization.

    BACKGROUND: Cellular senescence, a phenomenon of irreversible growth arrest of mammalian cells, is involved in various age-related phenomena in organisms. Hair follicle dermal papilla (DP) cells play important roles in the regulation of hair growth and loss. AIMS: We examined the implication of cellular senescence of DP cells in androgenetic alopecia (AGA), the most common form of male hair loss, and searched for the compounds that have a beneficial effect on the prevention of AGA. PATIENTS/METHODS: Expression of the 5α-reductase type II (SRD5A2) gene, which plays a key role in the development of AGA, was examined by quantitative RT-PCR and Western blotting analysis in DP cells. Besides, DP cells were cultured with the extracts of herbs used in traditional Ayurvedic medicine to search for the compounds that have a beneficial effect on the growth of DP cells. RESULTS: We found that expression of the SRD5A2 was up-regulated in senescent DP cells. We also found that the herbal extract of Plumbago zeylanica (root) enhanced the growth of DP cells and down-regulated the expression of SRD5A2 in DP cells. Further, plumbagin, an ingredient of P zeylanica, would be responsible for the above effects of P zeylanica. CONCLUSION: These results suggested the possibility that senescent DP cells may have a role in the development of AGA through up-regulating SRD5A2 expression, and the P zeylanica extract and plumbagin may suppress its development through enhancing the growth of DP cells and down-regulating SRD5A2 expression in DP cells.

    Cellular senescence is a phenomenon of irreversible growth arrest of mammalian somatic cells. Senescent cells increase the production of secretory proteins such as inflammatory cytokines, a phenomenon termed senescence-associated secretory phenotype (SASP). SASP is known to have profound effects on organismal health and aging; however, the molecular mechanisms of SASP are not precisely understood. In our previous studies, we have shown that senescent cells show decreased function of lamin B receptor (LBR), a nuclear membrane protein that regulates heterochromatin organization. Here we examined the implication of LBR in the regulation of SASP because senescent cells show altered heterochromatin organization, which would affect gene expression. We found that knock-down of LBR up-regulated the expression of the SASP factors such as IL-6, IL-8, and MMP1 in HeLa cells, even though cellular senescence was not induced by LBR knock-down. Conversely, enforced expression of LBR suppressed their up-regulated expression in senescent cells induced by excess thymidine. Further, our gene expression profile analysis also showed that many secretory proteins were up-regulated by LBR knock-down. We then analyzed the regulatory mechanisms of the expression of SASP factors by LBR, and found that the promoters of these SASP factors associated with LBR in normally growing cells, but dissociated from it in senescent cells. Additionally, we found that enforced expression of LBR decreased the generation of cytoplasmic DNA, which could be involved in SASP, in senescent cells. These findings suggested that LBR would play crucial roles in the regulation of SASP.

    Cellular senescence is a terminal growth arrest phenomenon in mammalian cells. Coordinated regulation of protein synthesis and degradation is required to maintain protein homeostasis in cells; however, senescent cells exhibit decreased activity of the proteasome, a major cellular proteolytic machinery, with an accumulation of proteins. Indeed, we showed that MG132, a proteasome inhibitor, induced cellular senescence through an accumulation of proteins in human cells. We then investigated the mechanisms of cellular senescence induced by protein accumulation by treating cells with MG132. We found that lamin B receptor (LBR), a nuclear membrane protein that regulates heterochromatin organization, was mislocalized and down-regulated in cells on treatment with MG132. Importantly, enforced expression of LBR suppressed cellular senescence induced by MG132. We also showed that LBR was involved in the regulation of chromatin organization in senescent cells, and that endoplasmic reticulum stress and autophagy were likely to be involved in the mislocalization and down-regulation of LBR. These findings indicate that decreased LBR function was responsible for the induction of cellular senescence by MG132, and thus suggest that protein accumulation caused by inhibition of the proteasome induced cellular senescence probably through chromatin dysregulation in human cells.

    Cellular senescence is a phenomenon of irreversible growth arrest in mammalian somatic cells in culture. Various stresses induce cellular senescence and indeed, we have found that excess thymidine effectively induces cellular senescence in human cells. Further, many reports indicate the implication of chromatin proteins in cellular senescence. Here we analysed the role of lamin B receptor (LBR), a nuclear envelope protein that regulates heterochromatin organization, in cellular senescence induced by excess thymidine. We then found that the LBR protein was down-regulated and showed aberrant localization in cells upon induction of cellular senescence by excess thymidine. Additionally, we also found that knock-down of LBR facilitated the induction of cellular senescence by excess thymidine in cancerous HeLa cells, and importantly, it induced cellular senescence in normal human diploid fibroblast TIG-7 cells. These results suggested that decreased LBR function is involved in the induction of cellular senescence in human cells.

    OBJECTIVE: Keratinocytes are the predominant cell type in the epidermis and play key roles in epidermal function. Thus, identification of the compounds that regulate the growth of keratinocytes is of importance. Here we searched for such compounds from the herbs used in traditional medicine Ayurveda. METHODS: Human keratinocytes were cultured in the presence or absence of the herbal extracts for 2 weeks; the effect of the extracts on cell growth was determined by staining the cells with Coomassie brilliant blue. To detect the compounds that regulate the growth of keratinocytes, the herbal extracts were subjected to high-performance liquid chromatography (HPLC). RESULTS: We found that the extract of Emblica officinalis enhanced the growth of keratinocytes in culture. Further, we fractionated the extract of E. officinalis using HPLC and identified the fractions responsible for the enhanced growth of keratinocytes. CONCLUSION: The extract of E. officinalis enhanced the growth of human keratinocytes in culture. E. officinalis contains the compounds that would be beneficial for human skin health because enhanced growth of keratinocytes would promote wound healing.

    OBJECTIVE: Chronic exposure to ultraviolet (UV) radiation induces skin photoaging (premature skin aging). UV irradiation generates reactive oxygen species (ROS), which are shown to play a pivotal role in skin photoaging. Ayurveda is a holistic traditional medical system, and Chyawanprash is one of the most popular formulations in Ayurveda. Since maintenance of the function and appearance of skin is important, we examined whether Chyawanprash has a protective effect on skin photoaging. METHODS: To examine the effect of Chyawanprash on skin photoaging, hairless mice were administered with Chyawanprash in drinking water for 3 weeks, and then repeatedly exposed to ultraviolet light B (UVB) irradiation (225 or 450 mJ/cm2) to induce skin photoaging. To further examine the function of Chyawanprash, its effects were examined in cells cultured in vitro. Chyawanprash was added in culture medium, and examined for the effect on the growth of human keratinocytes, and for the ability to eliminate ROS which generated by paraquat (50 μmol/L) in HeLa cells. RESULTS: UVB irradiation caused symptoms such as rough skin, erythema, and edema on the skin in hairless mice, but administration of Chyawanprash relieved these symptoms. Further, Chyawanprash significantly suppressed epidermal thickening, a typical marker of skin photoaging, in mice. We then analyzed the effect of Chyawanprash in human cells in culture, and found that Chyawanprash enhanced the growth of human keratinocytes, and efficiently eliminated ROS, which are causally involved in skin photoaging, in HeLa cells. CONCLUSION: These findings suggested that Chyawanprash may have beneficial effects on slowing skin photoaging.

    <I>C. elegans rad-8</I> has been isolated as an ultraviolet radiation hypersensitive mutant<SUP>1)</SUP> and we have also found that this mutant is oxygen hypersensitive<SUP>2)</SUP>. Reactive Oxygen Species (ROS) as well as radiation cause cellular damages and result in a variety of diseases including cancer and aging. It is well known that most endogenous ROS are produced from mitochondria. Mitochondria are specialized organelles that produce ATP, regulate programmed cell death, or apoptosis and produce the ROS as a byproduct of energy metabolism.
    In order to know whether oxidative sensitivity of <I>rad-8</I> mutant is correlated with mitochondrial oxidative stress, we examined, 1) amount of supeoxide anion (O<SUB>2</SUB><SUP>-</SUP>) one of ROS, from mitochondria and 2) carbonylated protein as a marker of oxidative stress, 3) apoptosis at embryonic stage, 4) growth rate and 5) energy metabolism. Our results showed that the level of O<SUB>2</SUB><SUP>-</SUP> and the carbonylated protein in mitochondria produced by the O<SUB>2</SUB><SUP>-</SUP> significantly increased compared to N2 wild type. The apoptosis also increased. On the other hand, the energy metabolisms and growth rate of <I>rad-8</I> were lower compared to N2. Furthermore, the body size of <I>rad-8</I> at the adult stage was smaller than N2.
    These data suggested that the function of <I>rad-8</I> gene is related with mitochondria. It is assumed that the energy metabolic abnormality in <I>rad-8</I> mitochondria leads to increase of oxidative stress and apoptosis by the ROS over production.<BR>

    Oxygen is essential for aerobic organisms but it generates reactive oxygen species (ROS) in the metabolic pathways. However, the mechanisms underlying the generation and functions of ROS are not understood well. Here we analyzed the ROS-sensitive mutants of the nematode Caenorhabditis elegans to study the mechanisms of aging. We also identified the responsible genes for the ROS-sensitive mutants and analyzed their functions. Additionally, since excessive generation of ROS is harmful for organisms, we screened for the compounds that decrease the toxicity of ROS with human cells in culture.

    Oxygen is essential for aerobic organisms, but generates reactive oxygen species (ROS), which can cause cellular dysfunction that would accelerate aging and diseases. We have analyzed the genes involved in the generation or the quenching of ROS in the nematode Caenorhabditis elegans and the yeast Saccharomyces cerevisiae as model organisms. In this study, we have successfully identified the gene involved in the regulation of oxidative stress sensitivity in C. elegans and characterized the function of the gene, which confers oxidative stress resistance, when overexpressed, in S. cerevisiae. We also screened for bioactive compounds that show a protective effect on oxidative stress in C. elegans.

    酸素は生命活動に必須であるが、代謝の過程で、反応性の高い活性酸素種(O_2^-、H_2O_2、・OH等)を生成する。そして、それらが生体分子を攻撃し、老化など、種々の生命機能の劣化を引き起こす。活性酸素の消去に関わる遺伝子としては、スーパーオキシドジスムターゼ、カタラーゼ、グルタチオンペルオキシダーゼ等が知られているが、実際の活性酸素の発生/消去機構は複雑で、詳細は明らかになっていない。そこで、それらに関与する遺伝子を明らかにするために、線虫Caenorhabditis elgansや、酵母Saccha romyces cerevisiaeを用いて、様々な活性酸素感受性変異体を単離し、酸化ストレスと寿命の解析を行った。
    線虫変異体oxy-4は、高濃度酸素下(60-90%酸素)で生育出来ず、活性酸素放出剤であるパラコートに対しても高い感受性を示す。寿命は野性型よりも短い。この変異体を詳細に解析し、oxy-4遺伝子を3番染色体上の非常に狭い領域にマップした。そこで、その領域に存在するDNAを、マイクロインジェクション法により変異体に導入したところ、1種類のDNAクローンが活性酸素感受性をほぼ完全に相補することを見いだした。しかし、このDNAを断片化し作成したコンストラクトは、十分な相補活性を示さなかった。さらに複数のコンストラクトを作成し実験を行ったが、領域を狭める事は出来なかった。そこで、領域内の遺伝子内にフレームシフト変異を導入し、原因遺伝子の同定を行い、原因遺伝子を同定する事に成功した。
    酵母に酸化ストレス耐性を付与出来る遺伝子として3種類の酵母遺伝子を単離した。SODやカタラーゼ、グルタチオンペルオキシダーゼ等の発現を調べたが、発現の上昇は観察されなかったので、本年は、上記抗酸化酵素の遺伝子を欠損させた株を作成し、これら遺伝子が、抗酸化酵素欠損株のバックグラウンドでも酸化ストレス耐性化能を示すか調べた。

    酸素は生命活動に必須であるが、、様々なストレスを引き起こす危険因子でもある。酸素は生体内で反応性の高い活性酸素(O_2-、H_2O_2、・OH等)を生成し、それらが生体分子を攻撃する。現在、活性酸素は老化の促進を引き起こす主要因子の一つと考えられている、本研究は、活性酸素と老化の関係を遺伝学的に明らかにするため、遺伝的解析の可能な線虫(Caenorhabditis elegans)を用いて、酸化ストレス・老化に関わる遺伝子を同定することを目標とする。戦略として、既に得られている長寿命/活性酸素耐性変異体(mev-4)、短寿命/活性酸素高感受性変異体(oxy-4)の原因遺伝子を期間内にクローン化する。
    変異体の原因遺伝子のマッピング、及び、同定
    mev-4の原因遺伝子は染色体Vにマッピングされた。様々な三因子交雑を繰り返し、詳細なマッピングを行った結果、mev-4はsma-1とlon-3の間にマッピングされた。その中に含まれる遺伝子を調べ、そのうちの一つに、変異が起きていることを見いだした。
    oxy-4の原因遺伝子についても、同様の手法を用いて、染色体IIIのdpy-17とlon-1の間にマップした。oxy-4の場合も、その領域に含まれる遺伝子に変異を起こしている遺伝子を見いだした。
    現在これらの遺伝子がmev-4、及び、oxy-4にこれら遺伝子を導入し、その表現型を相補するかどうか調べている。

    酸素は生命活動に必須であるが、様々なストレスを引き起こす危険因子でもある。酸素は生体内で反応性の高い活性酸素(O2-、H2O2、・OH等)を生成し、それらが生体分子を攻撃することによって、各種成人病や老化の促進を引き起こすと考えられている。生物は酸素障害から免れるため、活性酸素を消去する仕組みをもち、また活性酸素によって損傷した生体分子を修復する機構を備えている。本研究は、活性酸素に対する防御機構を遺伝生化学的に明らかにするため、遺伝的解析の可能な線虫(Caenorhabditis elegans)を用いて、活性酸素の防御機構に関わる遺伝子を同定することを目標とする。
    線虫(C.elegans)を変異源処理し、活性酸素耐性変異体を5種類、高感受性変異体を7種類単離した。全ての変異体について、可視マーカー変異体との標準的な掛け合わせにより、原因遺伝子の座位する染色体を同定した。さらに、同定された染色体上に存在する別の可視マーカー変異体と、掛け合わせを行うことにより、原因遺伝子座を絞り込んだ(3因子交雑)。
    このような実験を複数のマーカー変異体を用いて繰り返し、ほぼ全ての変異体に関して原因遺伝子のマッピングを終了した。そこで、絞り込まれた染色体領域に存在するコスミドを変異体に導入する実験を開始した。現在のところ、相補活性を持つコスミドの同定には至っていないが、原因遺伝子の同定は間近であると期待している。

    Approximately 50% of immortal human cell lines so far tested seem to have a mutation on a gene on human chromosome 7 since introduction of this chromosome by microcell-mediated chromosome transfer specifically suppress their division potential. To identify it, we prepared a panel of radiation hybrids which contain a small segment of human chromosome 7 under mouse cell background. By chromosome transfer and PCR with STS markers, we narrowed down the gene within a 2-3 Mb region of the chromosome. We planned to rescue human DNA form the hybrids with a newly developed TAR (Transformation Associated Recombination) cloning system in yeast. In collaboration with NIH that invented the cloning system, we prepared a YAC/BAC library from one radiation hybrid. These BAC clones were introduced to human Met5A cell line to examine their anti-proliferative activity against this cell line.
    As by-products of this study, we have shown that the immortality suppresser gene negatively regulates the telomere maintenance mechanisms in particular human cell lines. Upon introduction of the gene into the telomerase-positive lines, the telomerase catalytic subunit gene was down-regulated before entering senescence and with shortening of telomeres. In the negative lines, their usually long telomeres were immediately shortened.
